The [NiFe] hydrogenases from both bacteria have four metal ion binding sites. The binding sites for H(2) and a combination of Fe(3+) and Mg(2+) have been previously determined, whereas the Fe(2+) and Ni(2+) binding sites were revealed by our mutational studies and correspond to two different metal ion binding sites in the D. desulfuricans hydrogenase. A comparison of sequence alignments between Synergistes ureaseus and D. desulfuricans shows that Synergistes possesses two more metal ion binding sites in the [NiFe] hydrogenase. The sites are located on the surface of the ferredoxin domain of the [NiFe] hydrogenase from Synergistes, whereas the corresponding residues of the D. desulfuricans hydrogenase contain mutations, which result in loss of the metal ion binding specificity for H(2).The invention relates to a method of detecting pollutants of exhaust gas from internal combustion engines.
